A linear graph for digoxin radioimmunoassay.

چکیده
The determination of drug or hormone concentrations by radioimmunoassay involves interpolation of values for radioisotope counts within standard curves, a technique which requires some dexterity in curve drawing and which results in some inaccuracy in practice. A number of linear transformations (Ekins, 1974) and computer programs (Burger et al, 1972; Duddleson et al, 1972; Livesey, 1974) for standard curves which overcome this difficulty have been described, but most of these are complex and consequently time-consuming for routine use. In radioi'munoassays involving saturation of the antibody-binding sites a special case exists in that the bound radioactivity is directly proportional to the specific activity of ligand in the system. Thus a graph of the ratio of radioactivity bound in the absence to that in the presence of added nonradioactive ligand is linear against the concentration of added ligand (Hales and Randle, 1963). The present work, which supplements Hales and Randle's method, describes a modified expression of this relationship which has been applied to the determination of digoxin using a commercial kit. Specially constructed graph paper, which yields linearity with standard solutions, can be used directly without data transformation.

Microencapsulated antibodies in radioimmunoassay--I. Determination of digoxin.
We describe the application of the microencapsulated-antibody technique to the radioimmunoassay of digoxin in serum. Droplets of emulsified rabbit antibody are microencapsulated in a semipermeable nylon membrane by an interfacial polymerization technique. The antibody microcapsules are incubated with 125I-labeled digoxin and unlabeled digoxin for 15 min at 37 degrees C, then free and bound digo...
متن کاملAdaptation of the EMIT serum digoxin assay to a mini-disc centrifugal analyzer.
We have modified the EMIT (Syva) serum digoxin assay for use with a mini-disc centrifugal analyzer. A minicomputer is used to control data acquisition, calculations, and readout for 14 cuvettes simultaneously. Thus, it is not necessary for time each individual assay manually or to plot calibration curves on graph paper. We use incubation times and dilutions similar to those used for the manual ...
متن کاملMeasurement of plasma digoxin concentration by radioimmunoassay.
A rapid, sensitive, and precise method for measuring the plasma digoxin concentration has been developed with the radioimmunoassay technique. Seventy patients receiving digoxin were shown to have plasma digoxin concentrations between 0.4 and 5 ng./ml. Preliminary studies show that though there is a positive correlation between total daily dose and the plasma digoxin concentration, the relations...

the treatment of congestive heart failure and atrial fibrillation. It is important to measure digoxin in serum from digitalized patients because the therapeutic range of serum digoxin concentration is narrow. Immunoassays are routinely used for therapeutic drug monitoring (TDM) of digoxin. The antibody with cross-reactivity to metabolites of digoxin (digoxigenin bis and monodigitoxosides, digox...
متن کاملRadioimmunoassay of digoxin: an intercomparison of results with three methods.
The radioimmunoassay of digoxin has been examined by comparing RIA data obtained by a clinical laboratory using the method of Smith et al. [New Engi. J. Med. 281, 1212 (1969)] with data obtained in a laboratory using commercially produced [3H]digoxin and [1251]digoxin kits. Correlations between clinical laboratory data and the results with each kit and correlation between kits are presented. Th...
